Biosynthesis, partial purification and characterization of invertase through Carrot (Daucus carota L.) Peels

چکیده

The current work was investigated on the production of Invertase under solidstate fermentation (SSF) through Aspergillus species, by utilizing peels of Carrot (Daucus carota L.) as a substrate. The maximum production of Invertase (7.95±0.1 U/ml) was attained by using Aspergillus niger with 1×106 spores/ml at 90% moisture content after incubation period of 72 h. Through ammonium sulphate precipitation the enzyme was purified about 1.42-fold. Invertase showed maximum thermal stability ranges from 20-70°C at a pH range (5.5 to 6.5) with utmost activity at pH 5.5 and 50°C. However, the enzyme was highly active at two different concentrations of sucrose that is, 0.1 and 0.5 M respectively, but it indicates less activity at glycerol. While the enzyme was completely repressed by Hg2+ (1 mM) and slightly enthused by Co2+ and Na+ at the similar application.

Purification and characterization of neutral and alkaline invertase from carrot.

Neutral and alkaline invertase were identified in cells of a suspension culture of carrot (Daucus carota L.) and purified to electrophoretic homogeneity. Neutral invertase is an octamer with a molecular mass of 456 kD and subunits of 57 kD, whereas alkaline invertase is a tetramer with a molecular mass of 504 kD and subunits of 126 kD. Both enzymes had sharp pH profiles, with maximal activities...

Molecular characterization and functional analysis of sucrose-cleaving enzymes in carrot (Daucus carota L.).

The amount of carbon transported into storage organs of crop plants to a large degree determines crop yield. The role of sucrose-cleaving enzymes in this process is not clear and it is the main goal of our work to tackle this question. Sucrose cleavage is catalysed either by invertase or sucrose synthase both of which exist in several isoforms with different subcellular locations. Carrot (Daucu...

Partial purification, characterization, and thermal and high-pressure inactivation of pectin methylesterase from carrots (Daucus carrota L.).

Pectin methylesterase (PME) from carrots (Daucus carrota L.) was extracted and purified by affinity chromatography on a CNBr-Sepharose 4B-PME inhibitor column. A single protein and PME activity peak was obtained. A biochemical characterization in terms of molar mass (MM), isoelectric points (pI), and kinetic parameters of carrot PME was performed. In a second step, the thermal and high-pressure...

Isolation and characterization of a novel antifreeze protein from carrot (Daucus carota).

A modified assay for inhibition of ice recrystallization which allows unequivocal identification of activity in plant extracts is described. Using this assay a novel, cold-induced, 36 kDa antifreeze protein has been isolated from the tap root of cold-acclimated carrot (Daucus carota) plants. This protein inhibits the recrystallization of ice and exhibits thermal-hysteresis activity. The polypep...

Zeolite alleviates defense responses in drought stressed carrot (Daucus carota L.)

Drought stress is one of the main restrictions in plant production in arid and semi-arid regions. Adding superabsorbent agents that maintain water in the soil, is among strategies to cope with drought stress. Therefore, in order to investigate the effect of zeolite superabsorbent on the physiological properties of carrot plants, the experiment was carried out as a factorial in a completely rand...
